The Plasmid View

The plasmid View in Sesame allows the user to record information on plasmid constructs synthesized as part of a project. The plasmid record contains information on the physical characteristics and location of the construct. Plasmid records can be created and manipulated in the same ways as primer records. The plasmid records contains a few fields worth noting. The Construct field allows the user to describe key modules within the plasmid. These include affinity tags, protease sites and protein elements. To add a line to the construct field, click the green icon found on the right hand side of the field. Highlighting a line and clicking the red icon deletes the selected row. The Genetic Code field will accept a DNA sequence. Selecting Calculate AA Sequence from the Plasmids menu translates that genetic sequence into an amino acid sequence and deposits it into the AA Code field (note that the cursor must be placed in the AA Code field prior to asking Sesame to translate the DNA sequence into a protein sequence). The contents of this field are propagated to any protein record that is linked to the plasmid entry (if the user so chooses). Clicking on the purple icon to the left automatically calculates a few key physical characteristics for the peptide sequence.

As with all other record types, a plasmid record can be linked to other record types to create an electronic papertrail. Details can be found on the Linking Records page.
