Manipulating Work Groups |
|
| Whenever adding or deleting ORFs, the work group must be in an editable form. When a new work group is created, it is
automatically "locked", meaning that the Save Work Group command will not overwrite the ORF list in
the database. In order to add or delete ORFs the user must first select Unlock Orf List from the
Edit menu. To avoid accidental changes to a work group it is a good idea to relock the work group after modifying the ORF
list. After changing the ORF list, make sure that you save the work group.

| ORFs can be added to a work group in two ways. One or more ORFs can be easily transferred from the ORF view into a work group
where the user can then begin to track progress towards the final product, be it a plasmid, purified protein or 3 dimensional structure.
To add ORFs to a work group,
highlight the desired ORFs in the ORF view and drag and drop them to the frame of the appropriate work group on the Board.
ORFs can also be added to a work group by highlighting them within the ORF panel of a work group record displayed on the
Board and dragging and dropping them onto the frame of another work group open of the Board (note that adding ORFs by this method
will create a link in the electronic paper trail in Sesame and will result in the display of the previous work group and position in the
Prev Coord column of the ORF Panel). The work group must be saved in order
to permanently add the ORFs to the work group. There are a few things to note with
this process. First, due to a quirk in the Java Runtime Environment, it may appear that the ORF selection is lost while it is
being dragged on to the work group record, however, this is not the case. Second, the focus of the windows will not change even when you have
dragged the cursor over the destination work group record and there may be no obvious change to the work group record, however, the
ORFs will have been added to the new record. ORFs that have been placed in a work group can be removed at any time
by highlighting them and selecting Delete ORF(s) from the Edit menu. |

|
| Highlighting an ORF and selecting Show Visual Plate from the Work Groups menu results in a new
window showing the physical location of the construct in 96 well array (or whatever plate format was choosen in the General Panel).
The reverse is also true, clicking on a specific well on the visual plate will result in the ORF located in that spot being highlighted in
the list of ORFs in the Work Group View.

| In Sesame, it is possible to create variants of work groups, allowing users to track the progress of experiments running
in parallel or series to an orginal work group. To create a variant, open the work group that is to be used as the template and
select Save as New Variant from the Work Groups menu. A new record is automatically created and issued
a unique database identifier (note that for all ORFs, the Prev Coord column contains the work group number of the parent work group).
All fields in the general panel are automatically copied from the parent, with the exception of the mages and files associated with
the template work group. By default, all ORFs making up the parental work groups are copied to the variant. However, by highlighting
a subset of ORFs prior to selecting Save as New Variant, users can specify that only those particular ORFs are placed into the new variant. |
